The association of BRAF v600e mutations in the feces of colorectal cancer patients with location, histopathology, and tumor differentiation degree in Makassar, Indonesia

Abstract
Introduction: BRAF mutations (v-RAF murine sarcoma homogeneous viral oncogene B1) has been associated with aggressive phenotypes and are the main prognostic biomarkers for poor outcomes, especially in late-stage colorectal cancer (CRC). Faecal DNA testing has become a valid and reliable non-invasive procedure for the detection of CRC. This study aims to determine the relationship of the BRAF V600E mutation in CRC patients' faecal with the location, histopathology, and degree of tumour differentiation.
Material and Methods: This study used data from 43 stool samples of CRC patients at Wahidin Sudirohusodo Hospital Makassar from July to December 2019. We collected data such as age, sex, CRC location, histopathological results, degree of differentiation, and BRAF mutations. Statistical tests were performed with a Chi-squared test. The test results are significant if the P-value <0.05.
Results: There were 43 samples aged 21 to 80 years, with an average of 56.0 ± 11.2 years. There were 46.5% men and 53.5% women with a male-to-female ratio of 1:1.15. Most of the tumours are located in the right colon 18 (41.9%), rectum 14 (32.6%), and left colon 11 (25.6%). Non-mucinous adenocarcinoma is a major histopathological type compared to mucinous adenocarcinoma with 22 (51.2%) patients and 21 (48.8%) patients, respectively. There were 19 poorly differentiated tumours (44.2%), 15 moderately differentiated (34.9%), and 9 well-differentiated (20.9%). The BRAF V600E mutation was 6 (14%) and not the BRAF V600E 37 mutation (86.0%). BRAF V600E mutations were significantly related to tumour location, degree of differentiation, and histopathology (p <0.01). There was no significant relationship between sex and age.
Conclusion: There is a significant relationship between BRAF V600E mutations in CRC patients' faecal patients with location, histopathology, and the degree of tumour differentiation.
